The Tigers used six of their initial seven first-round picks on Wednesday night, headed by top selection Sam Lalor.
But they traded the last of them away, giving pick 27 to North Melbourne in a deal that landed them an extra first-round selection next year.
Now Richmond are listening to offers for pick 28 - the first selection on Thursday night - in a bid to further bolster their hand.
It comes as the Tigers attempt to turbo-charge their rebuild after finishing last in Adem Yze's first season as coach and losing several experienced players to trades, free agency and retirement.
"It was a pretty big night for the club," Richmond talent manager Blair Hartley said after Wednesday's first round.

"We had a pretty intense trade period which put us in the position where we could look to start what's next for the football club, and we're very excited."
Hartley said his phone was "running hot" towards the end of the first round, with North Melbourne eventually tempting the Tigers into a deal in order to secure key forward Matt Whitlock with the final pick.
Whitlock's twin brother, Jack, was one of the hard-luck stories on night one of the draft when left on the board, but is set to be picked when the draft resumes on Thursday night.
Richmond currently hold the first pick of night two but will listen to trade offers that would further strengthen their hand this year or next.
"That was part of our discussion even leading into the draft," Hartley said.
"We really rated (the depth of talent) but if we could maximise our position for next year as well and it worked for us, we'd look at it.
"We'll assess everything (in regards to pick 28). There's options in trading back as well, and there's still some pretty handy players on the board.
"So we're excited to see how the next 24 hours play out.
"It's a non-stop business, this one. I'd love to go and have a beer but I think the phone will still be running pretty hot. We've already had a couple of calls."
Lalor was joined at the Tigers by fellow midfielder Josh Smillie (pick seven), small forward Taj Hotton (12), key forwards Jonty Faull (14) and Harry Armstrong (23), and intercept defender Luke Trainor (21).

An explosive goal-kicking midfielder, Lalor attracted attention at junior level with his powerful frame and knack of fending off opponents.
The 18-year-old has understandably drawn comparisons to Tigers icon Dustin Martin and is even keen to take on the retired legend's No.4 jumper.
But Hartley, whose recruiting team watched all of Lalor's underage games this season, wants the new recruit to make his own name.
"I see Sam Lalor in the highlights. He's his own man," Hartley said.
"He's really strong-willed, he's got a great appetite for the contest and I'm not big on comparisons.
"I'd love for Sam to start from the beginning and work his way up and forge his own career."
