Halfback Reynolds was cleared of any serious neck injury in last Thursday night's 24-16 loss to Melbourne after he was taken from the field at AAMI Park on a medicab.
He was ruled to also have sustained a category two concussion and will need to go through NRL protocols to take his place against the Panthers on Thursday night at Suncorp Stadium, which is highly unlikely.
Reynolds trained on Tuesday away from the main group but the Broncos are set to err on the side of caution.
Former Wests Tigers half Madden filled in for regular No.6 Ezra Mam when he was suspended against South Sydney in round nine.
Madden had an unhappy night on that occasion in a 32-6 loss but he is a natural No.7 and Mam said he was backing him to fire.
"If Reyno' doesn't end up playing Jock will be stepping up," Mam said
"I am happy to take the lead but Jock is there as well and he will take the lead as well. He has played a lot of seven growing up."
The second-placed Broncos beat the third-placed Panthers in Penrith in round one but the 2021 and 2022 premiers fired a warning shot to the rest of the competition with their 48-4 demolition of Sydney Roosters last week.
"We know we all have to be on the same page from the start," Broncos centre Kotoni Staggs said.
"If we can play our shapes and defend in the middle as a team then I think we can go close to beating them. It is all about us.
"We don't need to worry about Penrith. We know how good Penrith is."
