At Sydney's Downing Centre Local Court on Thursday, Daniel John Keneally's lawyer Paul McGirr mentioned the not guilty plea.
The 24-year-old constable did not appear in court.
Attached to the Central Metropolitan Region, the Newtown man was changed in October of fabricating false evidence with intent to mislead a judicial tribunal.
On February 24, 2021, Keneally allegedly created a statement about a chat he had with Luke Moore. Details of the discussion were untrue, court documents claim.
Talking to reporters outside court, Mr McGirr declined to comment on what he called a "sensitive matter". He noted his client would fight the charge, saying he would assess a brief of evidence to be served by December 15.
The incident led to an investigation by the Law Enforcement Conduct Commission and the officer's arrest at Newtown Police Station.
Once the criminal proceedings are concluded, the LECC will provide a report to the NSW parliament.
After a review by NSW Police, Keneally has been suspended with pay.
