And the time has come once again for over 300 people to hit the road for the annual drive, which will stop off in neighbouring Wangaratta for a laugh-out-loud comedy night.
Bec Daniher, daughter of Neale and Fight MND manager said it was important to take awareness of the disease outside the metro area.
"This disease is not city based, it's found everywhere," Ms Daniher said.
"And regional communities have always been a big supporter of Fight MND, so this is an opportunity to thank that support and continue to push out that awareness."
Since former Melbourne football star Neale was first diagnosed with the disease in 2014, Ms Daniher said the community support had been crucial to a largely underfunded area of research.
"Prior to Dad's diagnosis, we didn't know anything of the disease," Ms Daniher said.
"It was a big blow, but dad has remained positive and continued to fight against it.
"And our way of doing this was to fund raise and raise awareness.
"We've had a significant increase in funding and have brilliant scientists, who are working hard around the clock to find the answers."
Ms Daniher encouraged all who could attend the fundraising night in Wangaratta to do so, with comedian Dilruk Jayasinga set to get people laughing, while there will also be a gala auction on the night.
But Ms Daniher would also love to see as many people along the route as well, as a convoy of cars hit the highways of regional Victoria.
"We love to see people on route when we pass through community towns," she said.
"We have 320 people on the drive and 87 different cars.
"Our drivers have been fundraising all year. It really is a celebration."
