Eleven of the new cases are linked to current outbreaks and eight have been in quarantine throughout their entire infectious period.
The source of four cases is still under investigation by health authorities.
Four cases are linked to Al-Taqwa College, two are linked to the Hobsons Bay outbreak, one is linked to the Caroline Springs shopping centre, one is a household contact of a City of Melbourne case and one is a household contact of a Brunswick case.
Two cases are linked to Glenroy West Primary School and one case is a household contact of a Glenroy case announced yesterday.
The final three unlinked case are a Wyndhamvale truck driver, a Bundoora worker who lives in Roxburgh park and a Middle Park resident.
Exposure sites have swelled above 400 overnight including the busy South Melbourne Market and McDonalds Tullamarine but none have been added in regional Victoria since the latest Delta outbreak began.
The cases were detected from 40,737 test results, with a record 27,427 vaccine doses administered across state-run sites.
With an "overwhelming" response to expanded vaccine eligibility at nine state-run sites, Premier Daniel Andrews has announced the availability of the AstraZeneca vaccine to 18 to 39-year olds will be extended across Victoria's 50 state-run vaccination hubs, with the establishment of a further three drive-through clinics.
It forms part of an ambitious plan to achieve one million jabs over the next five weeks.
"That is a significant undertaking but I'm very confident we will meet that target...a lot of the hesitancy I think is talk," Mr Andrews said.
"This is all about making sure we get jabs in arms and all of us play our part in getting to the other side of this.
"We aim by the end of September to be at that 60 per cent mark in state clinics...the quicker we get to 70 per cent, the quicker we get to 80 and then we're in a different world."
Chief Health Officer Brett Sutton said four unexpected wastewater detections of COVID-19 had been found in Camberwell, Balwyn, the western suburbs and Lakes Entrance, and urged anyone eligible to get vaccinated.
"The threat in Australia is greater than at any point since March last year," he said.
There were no new cases detected in hotel quarantine.
There are two cases in hospital with COVID-19 and one person in intensive care.
The news comes with 390 cases of community transmission acquired in NSW to 8pm last night, with "at least" 60 cases active in the community while infectious with the virus.
There have been two further COVID-19 related deaths - an unvaccinated woman in her 40s who passed away at home and a vaccinated man in his 90s who was in palliative care.
