After Ireland were bowled out for 286 on the second morning, Mahmudul put the tourists' bowlers to the sword as Bangladesh closed on 1-338, giving them a 52-run lead with nine wickets still standing.
Mahmudul and fellow opener Shadman Islam (80) put on 168 for the first wicket at the Sylhet Stadium on Wednesday, with Mominul Haque's unbeaten 80 then helping add another 170 runs without further loss before the close.
Matthew Humphreys took Ireland's solitary wicket, with Shadman caught behind by Lorcan Tucker in the 42nd over.
Mahmudul's career-best knock, which has so far occupied 283 balls, featured 14 fours and four sixes. Alarmingly, for Ireland, he had looked more composed the further the innings lasted and he will be fancying at least a big double-century when he resumes on Thursday.
Ireland had resumed on 8-270, but opener Paul Stirling failed to add to his overnight 60 and the tourists were only able to add another 16 runs.
